Wigan came from behind to inflict Hull KR's first defeat of the Super League campaign.
Trailing 12-6 at the break after Zach Eckersley's try, Warriors took control in the second half.
Jake Wardle, Harry Smith, Jai Field and Abbas Miski went over to send Wigan second in the early-season table, two points behind Hull KR.
The hosts looked well placed to make it seven straight league wins with tries from Joe Burgess and Peta Hiku but failed to score a point in the second half.
The Robins have seldom tasted defeat in the past year, winning 24 from 29 matches but four of those defeats have been by Wigan.
Hull KR came into the match off the back of a Challenge Cup quarter-final win at rivals Hull FC.
But it came at a cost with influential half-back Mikey Lewis absent with a groin injury he picked up in the derby game.
After Burgess dived into the left corner to score Hull KR's first try, Wigan replied as Bevan French offloaded to give Eckersley a 70-metre run-in.
Rovers were back ahead before the break as they kept the ball alive on the fourth tackle, ran the ball left and a short pass sent Hiku over.
Thereafter, it was all Warriors and Smith stepped off his right foot to score and put them ahead before French picked up a loose ball on halfway and offloaded to Field to run in and end the Robins' fine start to 2025.
Both sides face derby games in their next round of fixtures on Good Friday, 18 April.
Hull KR visit Hull FC and Wigan host St Helens, who are third after their 26-14 win over Wakefield.
Hull KR head coach Willie Peters told BBC Radio Humberside:
"I thought we put in a great effort, competed hard and were in control for a long time.
"I don't think the score reflected how tight the game was but it was there to be won.
"The difference between the two teams is that they're very good at taking their opportunities."
Wigan head coach Matt Peet told BBC Radio Manchester:
"We've beaten a good team here tonight.
"We were second best for large parts of the game and it took us a while to get to grips with it.
"But we'll always back our commitment to each other."
